Research on Quantitation of Almond Protein in Almond Protein Beverage Abstract：The aim of the research was to develop competitive enzyme linked immunosorbent assay (ELISA) for quantitation of almond protein in almond protein beverage. We applied two-dimensional electrophoresis (2-DE) to compare the differences between almond and apricot kernel to search for almond-specific proteins. A Pru-1 subunit fragment with a mass of 27 kDa and pI (isoelectric point) of 5.5-8.0 was found, extracted, purified and used as antigen to prepare a specific monoclonal antibody. Using the monoclonal antibody and almond soluble protein as standard, an optimized ELISA method was established with IC50 of 3.3 μg/mL and linear detection range of 5.0 to 200 μg/mL (R2=0.99). The assay did not register cross-reactivity with tested apricot kernels and other edible plant seeds. The sample extracting solution was 7M urea with 0.5%DTT. The limit of detection (LOD) was 100 μg/g, with relative standard deviation less than 10%. The average recovery rates were 97%, 93%and 84% under the sterilization condition of pasteurization, ultra high temperature treated and autoclaved sterilization, respectively.
