Another issue that needs to be explained is that the PHBVPEGMAL
surface (Figure 3c) does not suppress the cell growth
significantly as we expected because of the presence of PEG if comparedwithPHBV-
NH2 and PHBV-RGDsurfaces (Figure 3a,d). The
reason is likely to be the reactive maleimide groups at the end of PEG.
The maleimide group reacts specifically with sulfhydryl groups when
the pHof the reaction mixture is between pH 6.5 and 7.5 and forms a
stable thioether linkage. Because sulfhydryls are present in most
proteins and the cell culturemedium containsmany proteins, it is very
possible that the proteins in the culturemedium are chemically bound
to the end of PEG by maleimide groups, further promoting cell
adhesion and proliferation.

另一个需要加以说明的问题,如我们所料,与PHBVNH2和PHBV-RGD表面(图3a,d)相比较,PHBVPEGMAL表面(图3c)由于PEG的存在不会明显抑制细胞生长(图3a,d)。这可能是由于在PEG末端的反应性的顺丁烯二酰亚胺基团的原因。反应混合物pH值处于6.5和7.5之间时,顺丁烯二酰亚胺基团与硫氢基进行特定的反应并形成一个稳定的硫醚键。 因为巯基存在于大部分蛋白质之中并且细胞培养基中含有大量蛋白质,完全可能的是培养基中的蛋白质通过顺丁烯二酰亚胺基团化学键合到PEG的末端,进一步地促进细胞粘附和繁殖。
