                                                摘  要

木质纤维素转化燃料乙醇是近年来的研究热点。本文研究了木质纤维素酶法生产燃料乙醇的工艺优化,重点在于提高酶解效率及降低纤维素酶成本,具体包括以下几点内容:
纤维素酶性能表征研究:通过测定各种纤维素酶和半纤维素酶的滤纸酶活,纤维二糖酶活和蛋白含量,对酶的性能进行比较。
多酶辅配优化酶解的研究:鉴于木质纤维素的复杂组成,研究调整酶解中酶的种类带来的酶解效果变化。在玉米芯和秸秆体系中,比较了两种纤维素酶酶解效果,研究了在纤维素酶和纤维二糖酶中加入果胶酶或木聚糖酶对纤维素和半纤维素水解效率的影响,以及不同木聚糖酶酶量对酶解效果的变化情况。实验结果表明:多酶辅配对酶解效果的影响受底物种类、所用酶种和酶量的影响。果胶酶比木聚糖酶效果更好,较少的果胶酶即可很大程度上提高半纤维素水解效率。
酶的回收:为了提高酶的使用效率,降低酶使用成本,本文对于酶的回收再利用进行了研究。主要讨论了膜分离,固定化和重吸附三种酶回收方法,并对于重吸附做了尝试性试验,在试验中得到了比较好的回收效果。

Abstract

Wood cellulose into fuel ethanol is the research focus in recent years. This paper studies the wood cellulose enzyme production of fuel ethanol process optimization, the focus is to improve efficiency and reduce enzyme cellulase costs, specifically including the following:
Cellulase Characterization Study: through a variety of cellulose and hemicellulose enzyme activity of filter paper, fiber and protein activity of sugar content, the comparison of the performance.
Fu optimize the allocation of more of the enzyme research: In view of the complexity of wood cellulose, examining the adjustment of the type of enzyme in the enzyme effect change. Corn cob and straw in the system, compared the effects of two cellulose enzyme, the research in the cellulose fibers and the sugar-added pectinase or xylanase on the hydrolysis of cellulose and hemicellulose efficiency Implications, as well as different xylanase enzyme hydrolysis of the effect of the changes. The results showed that: multi-fu match the effects of the enzyme substrate types, the enzyme used by the species and quantity of the impact. Pectinase than xylanase better, less pectinase to a large extent, improve the efficiency of semi-cellulose hydrolysis.
Of the recovery: In order to improve efficiency in the use of enzymes, the use of lower-cost, the paper for the recycling and reuse of a study. Discussed membrane separation, fixed and re-absorption of three methods of recycling and re-absorption to try to do the test, the pilot of the recovery has been relatively good results.
