形成碳氮键的方法及其在药物合成中的应用

摘要: C,N原子是构成生物的基本元素,而许多对生命体有着重要调节作用的天然产物及药物等其分子结构中都存在有碳氮键的连接.因此,研究关于碳氮键的形成方法,对于指导我们进行有药理活性的天然产物全合成及结构修饰和药物合成有着重大的意义. 形成碳氮键的方法有很多,本文C-N键形成的机理上来划分,共分为五种方法,包括:亲电取代反应,亲核取代反应,亲电加成反应,亲核加成反应,以及涉及氮宾历程的反应。并粗略的列举了近几年来这些方法在药物合成中或精细化工上的具体应用。

The C, N atom constitutes the biological fundamental element, but many pair of life bodies have the important control action natural products and the medicine and so on its molecular structure exist have the carbon nitrogen key's connection. Therefore, studies about the carbon nitrogen key's method of formation, regarding instructs us to carry on has the pharmacology active natural products complete synthesis and the texture finishing and the medicine synthesis has the significant significance. Forms the carbon nitrogen key's method to have many, the mechanism which this article C-N key forms comes up the division, altogether divides into five methods, including: Kisses the electricity substitution reaction, the nucleophilic substitution reaction, kisses the electricity addition reaction, the nucleophilic addition reaction, as well as involves the nitrogen guest course the response. And sketchy has enumerated in the last few years these methods in the medicine synthesis or the fine chemical concrete application. Study on Formation of Carbon and Nitrogen Bonds and Its Application in drug synthesis

Abstract: C atom and N atom are essential elements of organisms and carbon-nitrogen bonds exist in the molecular structure of natural extracts and drug which play an important regulating role in organisms. Therefore, research on the formation of carbon and nitrogen bonds has significant meaning for guiding us to conduct pharmacological active natural products total synthesis and its structure modification and drug synthesis.  There are a lot of methods for formation of carbon-nitrogen bonds. In this paper, based on the formation mechanism of C-N bonds of division, it was divided into five ways: electrophilic substitution reaction, nucleophilic substitution reaction, electrophilic addition, nucleophilic addition and reaction involving nitrene course. These methods’ application in drug synthesis or fine chemical industry in recent years had also been cited.
