Biological buffers such as MOPS allow the pH of an aqueous solution to remain constant while the concentration of
hydrogen ions present changes. These buffers have pKa values at or near physiological pH. Biological buffers are
non-toxic to cells, and are not absorbed through cell membranes. The concentration, temperature, and ionic
composition of the medium has minimal affect on the buffering capacity. These buffers are resistant to enzymatic
and nonenzymatic degradation furthermore, they are essentially transparent to visible and ultraviolet light. MOPS is
used for RNA electrophoresis in agarose. It is available in a free acid and sodium salt form, and works exceptionally
well for formaldehyde gels at a 20mM concentration. The free acid form must be adjusted to the working pH with an
appropriate base such as sodium hydroxide, potassium hydroxide, or tetramethylammonium hydroxide. MOPS 10X
liquid concentrate is prepared with diethylpyrocarbonate (DEPC) treated water, leaving the solution RNase free.
