The excessive amount of acid in solution , the H+ which is not combined with amidogen, anti-base titration with alkali, to dissolve puuzczrq between the content of acid of chitosanacid and the content used in titration, and can calculated the content of free amino group in chitosan  get the degree of deacetylation (DD).

Acid-base titration is the simplest method for determining the content of freein chitosan.  The basic free amino group in chitosan can be protonated quantitatively by acid, thus forming colloidal solution of chitosan. The excess acid in solution , i.e., the free H+ which is not bonded to  amino group, can be reversely titrated with base. The amount of acid bound to free amino group can be calculated from the difference of the amount of acid used to  dissolve chitosan and that of the base for titration, further obtaining the content of free amino group in chitosan as well as its deacetylation degree (DD).
