酸碱滴定法是最简单的一种测定壳聚糖中自由氨基含量的方法,壳聚糖的自由氨基呈碱性,可与酸定量地发生质子化,形成壳聚糖的胶体溶液.溶液中过量的酸,亦即没有与氨基结合的游离的H+,用碱反滴定,用于溶解壳聚糖的酸量与滴定用去的碱量之差,即可推算出壳聚糖自由氨基结合酸的量,从而计算出壳聚糖中自由氨基的含量,可得到其脱乙酰度(D.D.)。

Acid-base titration is the most simple method of determine the content of free amino group in chitosan, the free-chitosan was alkaline, can protonated quantitatively with acid, formed colloidal solution of chitosan. The excessive amount of acid in solution , the H+ which is not combined with amidogen, anti-base titration with alkali, to dissolve puuzczrq between the content of acid of chitosanacid and the content used in titration, and can calculated the content of free amino group in chitosan  get the degree of deacetylation (DD).

Acid-base titration is the simplest method for determining the content of freein chitosan.  The basic free amino group in chitosan can be protonated quantitatively by acid, thus forming colloidal solution of chitosan. The excess acid in solution , i.e., the free H+ which is not bonded to  amino group, can be reversely titrated with base. The amount of acid bound to free amino group can be calculated from the difference of the amount of acid used to  dissolve chitosan and that of the base for titration, further obtaining the content of free amino group in chitosan as well as its deacetylation degree (DD).

In solution excessive acid, i.e. with amino union's dissociation's H+, with the alkali back titration, has not used in difference which of the alkali quantity dissolves the shell polyose the sour quantity and the titrate uses, then calculated the shell polyose free amino ties the complex acids the quantity, thus calculates in the shell polyose the free amino content, may obtain it to escape the acetyl (D.D.).
