测定CYP酶活性时为什么底物浓度一定要选在Km值附近，不能理解，求解。 已有1人参与

| 如题，为什么一定要选在Km值附近呢。为什么不能只要在平台期之前就行？有什么优势吗？鉴于每次问问题都没人回答，这回就发10个，免得浪费。。。。。签到不容易啊 |

| 最近看到的文献这段可能是一个比较好的解释吧：concentrationsDuring the selection of CYP probe reactions, special care shouldbe taken with the substrate concentrations. In fact, it is possible thatmultiple CYP isoforms will be able to catalyze the same metabolictransformation if the substrate concentration is inappropriate and,in this case, the measured CYP activity via metabolite formationcould not reflect that of the presumed CYP. For this reason, select-ing a substrate concentration at or below the Michaelis–Mentenconstant (Km) established for the test system is recommended toactivate mainly the considered CYP pathway and maintain highselectivity.
