Abstract: In this study, we observed that 5′-end 6-mercapto-1-hexane-labeled single-stranded DNA (HS-ssDNA) dramatically enhanced the specificity of DNA duplication during the polymerase chain reaction (PCR). The regulation of PCR by HS-ssDNA is both intriguing and useful, and such findings further our understanding of the biochemical characteristics and potential applications of HS-ssDNA.
