This paper screened the best macroporous resin and its optimum conditions in the process of purifying total flavonoids in natural plant water smartweed (Polygonum hydropiper L) , used single-factor tests to examine the effects of type of resin, sample concentration, pH of the sample , the sample flow rate, eluent concentration, elution rate, dynamic adsorption resin, number of repeated use and other factors on purification.Using UV spectrophotometry to examine the content of total flavonoids, then calculated the amount of adsorption solution, parsing and extraction rate of total flavonoids content, finally through the validated tests to determine the optimum conditions are as followed:the type of resin is ADS-7, the sample solution concentration is 0.8856 mg / mL, the value of pH is 4.48, the resin sample volume is 5 mL / g; eluted with 50% ethanol, at a flow rate of 3 mL / min, eluent consumes 300 mL. After the treatment of ADS-7 resin,mass fraction of total flavonoids reaches 76.43%, the yield is 93.04%. Conclusion: ADS-7 macroporous resin does the best in the process of separation and purification of water smartweed flavonoids, it is an ideal medium in separation and purification.
