Use a column packed with octadecylsilane chemically bonded silica(4.6mm * 250mm,5μm),0.2mol/L sulfate buffer（prepared by dissolving 28.4g of anhydrous sodium sulfate in water and adding 2.7ml of phosphoric acid, adjusting the solution with ethanolamine to pH 2.3, diluting with water to 1000ml）and acetonitrile（74:26）as mobile phase with a flowrate 1ml /min, maintain the column temperature at 40℃,the detection wavelength is 214nm.Dilute a quantity of sample with 0.01mol/L HCl solution to produce a 40u/mL solution, inject 20μL into the column, record the chromatograms. Repeat the operation, using a quantity of insulin CRS.Calculate the units with respect to the peak area obtained in the chromatogram by the external standard method.
