构建酵母系统发育树问题-急，请大侠给予帮助已有2人参与

我用5.8S/ITS鉴定的酵母，我有90多株，建系统发育树时，没有把所有的菌都构建上面，差不多用了50株来构建树，参考菌株选的是blast最相近的菌株。审稿人主要对系统发育树一块提了很多问题。 1.The ITS sequence of each corresponding type strain should be added to the analysis, instead of the sequences which have been found in the literature。 参考菌株让我选择模式菌株，一定是标着模式菌株的菌株做参考才行吗，我找的带ACTT这样的菌可以吗 2.ITS may be quite variable between species. Thus, it is not the best marker for phylogenetic reconstruction. Here quite distantly related Saccharomycotina and Basidiomycota sequences are compared and aligned; I would like to know what exactly has been compared, because I suspect that the sequences of these species that can be aligned may be quite reduced and therefore poorly informative. 他说5.8S/ITS鉴定方法不是理想的方法,我并不是很懂，我就只用了这个方法鉴定，个别不够理想的结果才用了26S鉴定，但文章中我没有提26s，这个我应该怎样去回复审稿人呢 3.The work also lacks comparison between the (molecular or physiological) characteristics of the yeasts described and the reference i.e. the type-strains of the species found。审稿人让我分析比较一下我鉴定的菌和相应的模式菌株，两个审稿人都有提到这个问题。这个一般应该怎样去分析呢？在树里面他们是处于一个分支的，是说一下他们的相似度还是同源可靠性呢，求解， 还有一个审稿人说Yeasts used in the study were molecular identified using ITS-PCR method. Sometimes, a single method is not sufficient for the accurate identification of fungi. Molecular biological techniques are constantly in progress so yeast isolates must be classified on the basis of a polyphasic approach.文章中我用了生理生化去鉴定了一下，又用了一个5.8S/ITS分子鉴定，但这个审稿人说要多种方法结合，这也是对我用的5.8S/ITS不太相信吗？我还需要再加上一种鉴定方法吗？ 另外我想问一下，如果我用5.8s/ITS和26S，那我的系统发育树应该如何构建呢，是两个放一起构建一个树还是分开各建一个树呢。建树的时候要包含所有的菌还是取其中一部分建就可以呢？ 文章之给了一个月时间修改，分子鉴定这块并不是很懂，希望大家谁知道的能给我点帮助，哪怕一点点也好呢，不胜感激！金币较少，请大家见谅 |
