抗菌肽(Antimicrobial peptides)是一类具有抗菌活性的短肽,分子量在1000-7000左右,由12-50个氨基酸组成,具有光谱抗菌性,可杀死革兰氏阳性菌、革兰氏阴性菌、真菌等。抗菌肽ORBK是来源于无指盘臭娃Odorrcma grahami皮肤分泌物的含16个氨基酸残基的阳离子多肽。该多肽是一类环形肽,不仅具有抗菌活性,还具有稳定的促胰岛素释放活性。目前获得抗菌肽ORBK的方法只有提取和合成,从无指盘臭娃皮肤分泌物中提取存在提取量少的瓶颈,而化学合成法成本较高,因为该多肽合成中还需要一个环化的过程。在本论文中,我们利用基因工程结合H2O2氧化的方法,成功获得了抗菌肽ORBK,并对该抗菌肽的作用机制进行了探究,同时,我们还对抗结合杆菌多肽D29P63和PK34进行了初步研究。

Antimicrobial peptides  are antimicrobial  peptides  with  molecular weight of about 1000-7000 and consists  of 12-50 amino acids whose  antibacterial spectrum include  Gram-positive bacteria, Gram-negative bacteria and fungi. ORBK antimicrobial peptides are cationic peptides derived from  Odorrcma grahami skin secretions containing 16 amino acid residues. A  cyclic peptide, ORBK antimicrobial peptides have not only  antimicrobial activity , but also a stable insulinotropic activity. Currently  ORBK antimicrobial peptide can only  be extracted  and synthesized .  The much smaller yields of Odorrcma grahami skin secretions extraction is a critical technical bottleneck  and  the cost of chemical synthesis  is  high  because it  requires a cyclic process . In this paper, we use a genetic engineering   combined H2O2 oxidation method, successfully obtained  ORBK antimicrobial peptides .The   mechanism of antimicrobial peptides has been explored  and  bacteria binding peptides D29P63 and PK34 were studied  also .
