Block the 3' end of a DNA oligo

金币不多，求各位帮忙。 I want to irreversibly block the 3’ end of a DNA oligo so that it cannot be removed by the 3’-to-5’ exonuclease activity of proof-reading DNA polymerases (e.g. Platinum Pfx or Phusion Hot Start II). I have tried inverted dT, but it gets removed. Is there a way to block the 3’ end of a DNA oligo so it can withstand thermocycling in the presence of a proof-reading polymerase? Thanks, |
