纤维原料是地球上现今最丰富、可再生的廉价的重要资源,对它的应用的研究一直是科学界的重点和热点。纤维素酶是一种高活性生物催化剂。随着现代微生物技术的发展,纤维素酶在饲料、酒精、纺织和食品等领域显现出巨大的市场潜力。
        实验室课题组对双功能纤维素酶CbCel9A1CBM各模块功能研究后发现其各截短蛋白间存在渐进性和协同性。为进一步具体从结构-功能的关系了解各截短蛋白,我们希望能够对CbCel 9A-L和L-CBM3C模块蛋白经行晶体结构的解析,从而能从根本上了解其作用机理。首先,我们对目标蛋白经行了Ni-柱亲和分离纯化,并通过分子筛层析纯化发现纯化效果很好。之后根据SDS-PAGE凝胶电泳对目标蛋白的纯度进行了考证后用考马斯亮蓝法对目标蛋白的浓度进行了测定,并同时浓缩蛋白浓度。之后用悬滴式蒸汽扩散法进行大量实验后,确定了目标蛋白的最优结晶条件并进行了单晶培养。最后集中送往上海光源中国科学院上海应用物理研究所进行X-光同步辐射衍射解析晶体结构。

Many emphasis and attractions were paid to their applications  in this field. fiber enzyme is a kind of highly activated biocaltalyst. with the developement of modern microbiotechnology, giant maketing potentials of fiber catalysts have appeared in the fields of fodders, alcohol, textile and food.

很多专业名词不是很懂,抛砖引玉吧
