| 阿维菌素发酵生产过程中，伴随着浓重的气味物质—土味素（geosmin）产生，这不仅影响了阿维菌素本身的发酵代谢过程，而且降低了目标产物的合成效率，更严重的是对环境和人类日常生活产生极大的危害。然而,其代谢机制却很难单独从现有的基因学、酶学和发酵环境的理论角度进行阐明。本课题利用13C同位素标记技术研究土味素在阿维链霉菌中与阿维菌素生物合成之间的协同代谢机制。首先通过以特定标记的土味素代谢试验，探索菌体生长期和产物合成期土味素在胞内的分解代谢机制和代谢流向；其次，根据不同13C 标记的葡萄糖和土味素组合实验，探索土味素对生产菌呼吸代谢、中心碳代谢和阿维菌素合成代谢途径通量变化，揭示阿维链霉菌中土味素的合成机制和代谢网络调节机制，实现其宏观代谢和微观代谢研究相结合，对阿维菌素发酵代谢调控及工艺优化有着重要的理论和现实意义。 |

During the fermentation process of Avermectin, one substance “geosmin” with a strong odor was generated, which will not only affect the fermentation metabolism process itself, but also reduce the synthesis efficiency for the target product. More seriously, it will do a great harm to the environment and everyday life of our human beings. However, it is difficult to elucidate the metabolic mechanism separately from the theoretical point of view of genomics, enzymology and fermentation environment. In this paper, the collaborative metabolism for geosmin between the avermitilis and biosynthesis of avermectin has been investigated by using 13C isotopic labeling technique. Firstly, through the testing metabolic process of geosmin with specific marker, the mechanism of geosmin intracellular catabolism and metabolic flows in the cells during the period of bacterial growth and product synthesis has been explored. Secondly, based on the combination experiments of different 13C-labeled glucose and geosmin, the geosmin-producing bacteria respiration metabolism, central carbon metabolism and pathway flux of avermectin synthesis has been investigated. In this way, the synthesis mechanisms and metabolic network avermitilis adjustment mechanism for geosmin was achieved, resulting in the combination of macro metabolic and micro metabolism, which has important theoretical and practical significance to regulation and optimization the fermentation metabolic process of avermectin. 非本专业，纯属娱乐。  |
