During the fermentation process of Avermectin, one substance “geosmin” with a strong odor was generated, which will not only affect the fermentation metabolism process itself, but also reduce the synthesis efficiency for the target product. More seriously, it will do a great harm to the environment and everyday life of our human beings. However, it is difficult to elucidate the metabolic mechanism separately from the theoretical point of view of genomics, enzymology and fermentation environment. In this paper, the collaborative metabolism for geosmin between the avermitilis and biosynthesis of avermectin has been investigated by using 13C isotopic labeling technique. Firstly, through the testing metabolic process of geosmin with specific marker, the mechanism of geosmin intracellular catabolism and metabolic flows in the cells during the period of bacterial growth and product synthesis has been explored. Secondly, based on the combination experiments of different 13C-labeled glucose and geosmin, the geosmin-producing bacteria respiration metabolism, central carbon metabolism and pathway flux of avermectin synthesis has been investigated. In this way, the synthesis mechanisms and metabolic network avermitilis adjustment mechanism for geosmin was achieved, resulting in the combination of macro metabolic and micro metabolism, which has important theoretical and practical significance to regulation and optimization the fermentation metabolic process of avermectin.
