About soil sample pretreatment (molecular biology experts are welcomed)

Questions about soil sample pretreatment before total DNA extraction for PCR (qPCR or common PCR) (1) Drying: 80℃ or 110℃？ Why some literatures used 110℃？ The high temperature won't destory the DNA? (2) Enrichment in NB or NA and collection of the culture or colonies? (3) Dispersing the soil sample by glass beads to let the bacteria released? Which one is the most approperiate and most commonly used? I used the DNA for template of real-time quatitative PCR. [ Last edited by cashehe on 2012-8-6 at 20:46 ] |
