求助：审稿意见中有些不明白的地方，请教各位了

我的论文中只写了培养基，每个方法里面写了细菌培养好后离心洗涤的处理过程。但审稿人提出：How are the cultures done? Pre culture or main culture.Centrifugation process?? How many bacterial cells were counted?? 不知道该怎么回答，特别是里面的pre culture 和main culture是什么意思，请教各位了！ |

The sterile culture medium contained 5 g NaCl, 10 g peptone ( purchased from sigma ), 5 g yeast extract ( purchased from sigma ) per liter. The pH of medium was adjusted to 7.0 before autoclaving. It was sterilized in high pressure steam for 30 min at 121 ℃. |

这个问题人家是说你对培养物的制备说的太不详细了。Culture medium: 1) It lacks details: How are the cultures done? Pre culture or main culture.Centrifugation process?? How many bacterial cells were counted?? Just mentioning LB medium is not sufficient. 主要是缺少细节：培养物的如何得到的？培养条件楼主没提到，是多少温度，多少时间等等。种子培养还是发酵培养。离心过程的条件，包括转速、时间等。细胞计数的方法要涉及到。 |
