EXPERIMENTAL EXAMPLE 1-1: Inhibition of Degranulation of Mast Cells - β-Hexosaminidase Assay The rat basophilic leukemia cell line RBL-2H3 was seeded at a density of 2x105 cells/well in MEM supplemented with 15% FBS and 100 units/ml penicillin and streptomycin and incubated for 24 hours at 37ºC in a 5% CO2 atmosphere. After removal of the medium, the cells were incubated for 4 hours in a fresh MEM medium containing 25 ng/ml anti-DNP IgE. Then, the cells were washed with PIPES buffer (25mM PIPES, 119 mM NaCl, 5 mM KCl, 1 mM CaCl2, 0.4 mM MgCl2, 40 mM NaOH, 5.6 mM glucose, 0.1% BSA) before pre-incubation for 10 min. Thereafter, the cells were treated for 20 min with 25, 50 and 100 μg/ml the samples of Example 1 and then for 20 min with 50 ng/ml DNP-BSA. The reaction was terminated at 4ºC and the supernatant was plated in an amount of 25 μL/well into 96-well plates and reacted with 25 μL of 1 mM P-nitrophenyl-acetyl-β-D-glucosaminide at 37ºC for 1 hour in each well. The reaction was terminated with 0.1 M citrate buffer before absorbance at 405 nm was read in an ELISA reader. |

实验1-1：肥大细胞脱颗粒-β-氨基己糖苷酶的抑制试验 大鼠嗜碱性粒细胞系RBL-2H3种植到浓度为 2x105 个细胞/孔、含有15% FBS 和100单位/ml 青霉素和链霉素的MEM培养基中，37度下、5%co2环境中，培养24小时。移去培养介质后，细胞在含有25 ng/ml 抗体免疫球蛋白E的新MEM培养基中培养4小时。之后，在预培养前用 PIPES 缓冲液(25mg/ml PIPES, 119 mg/ml NaCl, 5 mg/ml KCl, 1 mg/ml CaCl2, 0.4 mg/ml MgCl2, 40 mg/ml NaOH, 5.6 mg/ml 葡萄糖, 0.1% 牛血清蛋白)洗涤细胞。随后，细胞用 25, 50 以及 100 μg/ml的实验1样品处理20分钟，再用50 ng/ml 抗体-牛血清蛋白处理20分钟。反应在4……后终止，上清液以25 μL/孔的量加入96-孔的培养皿中，每孔加入25 μL1mg/ml的对硝基苯酚-乙酰基-β-D-氨基葡萄糖苷与之在37……反应一小时。在用ELISA测定仪测定405nm处的吸光度前，用0.1mol/l的柠檬酸缓冲液终止反应。 有些乱码的自己添加上去  |
