Abstract: Microorganisms that can survive in extreme environments, demonstrate the adaptability of life to the harsh environments. The moderate halophilic bacteria in the Yuncheng saline lake just belong to this category. In the tests, the physiological and biochemical identification of W33 was achieved by enzyme assays by using oxidases, catalase, amylase enzymes, etc. The morphological analysis was also used to observe the cell morphology, gram stain of moderate halophilic bacteria. Finally, DNA extraction, PCR amplification and other methods were used to test the bacteria. W33 can grow in CML medium with a NaCl concentration range from 0 %~20 %，and a pH range from 5-10. The Gram stain results were positive.
