3.15 DNA与BCDO作用方式的研究 通过上述实验可知BCDO测定DNA的体系属于一种猝灭形式的体系,因此根据Stern-Volmer(即S-V)方程:F0/F = 1 + Kq [Q](式中:F0与F分别表示猝灭剂DNA不存在和存在时体系的荧光强度,[Q]表示猝灭剂DNA的浓度,Kq表示猝灭常数,单位为L•mol-1,其大小反映荧光探针与DNA分子之间的亲和力强弱),由图3.31拟合F0/F~CDNA图,结果如图3.32所示。 由图3.32可以看出,F0/F~CDNA有良好的线性关系,通过拟合,F0/F~CDNA方程为:F0/F = 1.05688 + 0.05808 CDNA,式中:CDNA的单位为mg•L-1,该DNA的F0/F~CDNA方程符合静态猝灭方程,该直线的斜率即是猝灭常数Kq,由于上述方程中DNA的浓度的单位为mg•L-1,依此对上述拟合方程Kq值进行单位换算,得出Kq大小为9.12×106 L•mol-1,这充分说明了本文中自制的新型荧光分子BCDO与DNA发生了强烈的相互作用,两者间有很强的亲和力。 |

3.15 The study of way DNA interact with BCDO Above experiments indicate that the system that BCDO determined DNA belongs to a quenching form system, according to the Stern-Volmer (S-V) equation: F0 / F = 1 + Kq [Q] (in the equation : F0 and F stand for fluorescence intensity of system without and with quenching agent DNA ,respectively.[Q] is the concentration of quenching agent DNA, Kq is quenching constant, its unit is L mol-1, and its size reflects the strength of affinity between fluorescence probe and DNA molecular , figure 3.31 fitting by F0 / F ~ CDNA figure, the results as shown in figure 3.32. AS the Figure 3.32 shows , F0 / F - CDNA has a good linear relationship, after fitted,the equation of F0 / F - CDNA is: F0 / F = 1.05688 + 0.05808 CDNA, in the equation: CDNA's unit is the Mg L-1, the F0 / F - CDNA equation of DNA agree with static quenching equation, the slope for the linear is the quenching constant Kq, due to the concentration unit of DNA in the above fitted equation is Mg L-1, according to the Kq value of above equation for unit conversion, the Kq equels 9.12 x 106 L, mol-1, this illustrated fully that in this paper a self-made new type fluorescent molecules such as BCDO had interacted strongly with DNA , and had a very strong affinity between them. 仅供参考,嘿嘿。 |
