The effects of DNA on BCDO's fluorescence spectra.
Under the better experimental condition, we studied the effects of DNA on BCDO's fluorescence spectra at seven calf thymus DNA concentrations, i.e., 0.0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0 (mg/L), and the results are shown in Figure 3.30. From figure 3.30, we see that as the concentration of DNA increases the peak position of BCDO's fluorescence emission spectra did not change at λEm=517.8nm, however, the fluorescence intensity decreased continuously (i.e., fluorescence quenching getting stronger). In our study system, the phenomenon that the addition of DNA decreased BCDO’s fluorescence intensity was consistent with the phenomena that fluorescence intensity decreases with the presence of DNA, the commonly accepted mechanism is that the fluorescent materials’ excited states become closer to Guanosine when certain small fluorescent molecules inserted into the base pairs of DNA, and Guanosine’s very strong electron-donating capacity makes electron transfer between the material and DNA bases. It is very likely that the conjugate plane of BCDO made by ourselves inserted into the base pairs inside of DNA double helix and makes the electron transfer between BCDO and Guanosine, and resulted in energy transfer failure and fluorescence quenching. |
