| 脂氧酶（LOX）为活性中心含铁的金属酶。本文将以大豆LOX为模板，研究外源金属离子对LOX活性中心的影响及其对酶活的调控，验证和完善我们提出的外源金属离子与LOX活性中心离子存在离子交换的假说，研究LOX催化过程中的电子传递/离子交换机制。利用Irving-Williams序列实现一些外源过度金属离子与LOX中铁的交换，研究其它金属离子对LOX的活性或酶促反应的影响；采用自制新型指示剂结合光谱技术在一定范围内定性定量地研究LOX活性的重构对其结构、活性及催化动力学的影响；实现对LOX的金属修饰并调控其催化动力学。本文是金属酶化工的研究和应用中亟需解决的共性问题，是对酶化工、天然酶修饰及金属离子生物效应等的应用基础研究；对了解原生态金属酶活性中心金属离子的作用机制、内外源金属离子间的相互作用及其活性调控具有普遍理论意义，也可为研究其它酶促反应中金属离子的作用机制提供方法上的参考。 |

LOL is a metalloenzyme with Fe in its active site.
