¹©²Î¿¼£º Gibberellic disease cause serious cuts in wheat production, while neither chemical control and prevention nor disease-resistant varieties succeeded in inhibiting it. By taking advantage of the actinomycete JK-1 polyketide found in our laboratory which has proved to be highly effective in inhibiting Fusarium graminearum, the present study, through preparation of probes using degenerate primer and obtaining of full-length PKS genes by constructing a genomic BAC library, completed the PKS gene expression in Escherichia coli and Agrobacterium-mediated transient expression of wheat, solved the technical difficulties in the transformation and expression of PKS genes in wheat, addressed the molecular mechanism of PKS gene¡¯s disease-resistant function, and clarified the significant role PKS genes played in preventing and controlling wheat scab. The project is expected to establish a set of techniques for the transformation and expression of PKS genes in wheat, revealing the molecular mechanism of the PKS gene¡¯s resistance to wheat scab. The research findings of the project shall shed light on obtaining wheat varieties with more stable and lasting resistance to wheat scab, compared with traditional breeding and transgenic disease-resistant wheat varieties. Implementation of the project scientific could provide evidence for the successful expression of PKS genes in plants and their strong resistance to diseases. |
