针对赤霉病导致小麦严重减产，而化学防治和抗病新品种均无法抑制赤霉病的危害问题，以本实验室发现的放线菌JK-1聚酮化合物能高效抑制赤霉病菌为契机，采用简并引物制备探针、构建基因组BAC文库获取PKS基因全长，并进行PKS基因的大肠杆菌表达及农杆菌介导的小麦转化表达，研究PKS基因在小麦中成功转化并表达的技术难题，探讨PKS基因在小麦的抗病作用分子机理，阐明PKS基因在小麦赤霉病防控方面的重大作用。项目预期将建立PKS基因转化小麦并成功表达的技术体系，揭示PKS基因抗赤霉病的分子机理。项目的研究结果对于获得比传统育种及转基因抗病小麦品种更稳定而持久的抗赤霉病小麦品种具有重要意义。项目的实施可以为PKS基因在植物上成功表达并表现出很好的抗病性提供科学依据。 [ Last edited by heihei3910 on 2012-3-5 at 22:11 ] |

供参考： Gibberellic disease cause serious cuts in wheat production, while neither chemical control and prevention nor disease-resistant varieties succeeded in inhibiting it. By taking advantage of the actinomycete JK-1 polyketide found in our laboratory which has proved to be highly effective in inhibiting Fusarium graminearum, the present study, through preparation of probes using degenerate primer and obtaining of full-length PKS genes by constructing a genomic BAC library, completed the PKS gene expression in Escherichia coli and Agrobacterium-mediated transient expression of wheat, solved the technical difficulties in the transformation and expression of PKS genes in wheat, addressed the molecular mechanism of PKS gene’s disease-resistant function, and clarified the significant role PKS genes played in preventing and controlling wheat scab. The project is expected to establish a set of techniques for the transformation and expression of PKS genes in wheat, revealing the molecular mechanism of the PKS gene’s resistance to wheat scab. The research findings of the project shall shed light on obtaining wheat varieties with more stable and lasting resistance to wheat scab, compared with traditional breeding and transgenic disease-resistant wheat varieties. Implementation of the project scientific could provide evidence for the successful expression of PKS genes in plants and their strong resistance to diseases. |

The implementation of the project for the PKS genes in plants express and show successful on very good resistance to provide the scientific basis. |
