When scanning by DMA through to higher temperatures the sample dries out and Tg estimations of the dried state can then be seen if the sample is subsequently reanalysed. Initial studies indicated that DMA may give useful data about the Tg of the lyophilized dry state, and this application could be inferred from other DMA studies performed on dry excipients [9]. This dry state Tg can be important in the prediction of the stability of solid dosage form in pharmaceutical products [4]. Lyophilized materials are hygroscopic and so controlled relative humidity (RH) would be vital when looking at these events as the water content would mark- edly affect the observed Tg. DMA has previously been used to investigate the glass transition of the dried state [17] in pharmaceutically relevant materials. Recently, a study has been published from Wu et al. [21] demonstrating the use of DMA to determine values for the Tg0 of trehalose along with mDSC and atomic force microscopy. They detected in the frozen liquid state the existence of two transitions at 35 and 28 C by mDSC and suggested that the former was associated with a glass transition and the second with a loss of crystallinity.
