Banna Mini-pig Inbred Line (BMI) is the first successful cultivated large mammal inbred line in the world, which is one of the major scientific and technological achievements with independent intellectual property rights innovated by Chinese primitively. It was found to be valuable for research and application in the bio-medical field such as xenotransplantation. In this research, main attempts had been taken to explore all the genes related to the immunological rejection in the xenotransplantation of BMI to human based on transcriptome sequencing technology (including unknown and rare transcription). The obtained data was processed by functional enrichment and cluster analysis to screen the major genes associated with xenotransplantation rejection. In order to determine the genes with high level expression in major organs for xenotransplantation, a multiple tissue expression in transcription levels was detected by real-time qRT-PCR, after which the function was further identified by prokaryotic and eukaryotic cells at protein expression level. Finally, the main exclusive genes related to major transplanted organs  were identifiedin at the DNA-RNA-protein level. All of these research has provided a background knowledge foundation for studying the genes correlated with immunological rejection in the xenotransplantation of pig to human. Besides, it also can provide the gene mapping framework for gene silencing, gene modification and gene knock as well as further detection of the important target genes. It could therefore lay the foundation for the BMI research and application in the xenotransplantation of pig to human field.
