请高手帮忙润色摘要,谢谢! 版纳微型猪近交系是世界上第一个培育成功的大型哺乳动物近交系,属于我国原始创新并拥有自主知识产权的重大科技成果,在异种器官移植等生命医学领域具有重大研究和应用价值。本研究拟通过转录组测序技术深度挖掘版纳微型猪近交系猪→人异种器官移植中与免疫排斥反应相关的所有基因(包括未知转录本和稀有转录本)。将获得的数据进行功能富集分析和聚类分析,筛选出与移植排斥反应相关的重要基因。采用实时荧光定量PCR技术进行多组织转录水平检测,确定在可用于移植的各主要脏器中表达丰度较高的基因,再进行蛋白水平的原核和真核表达方面的功能鉴定。最终从DNA—RNA—蛋白质水平确定出与各大脏器移植相关的主要排斥基因。为猪→人异种移植中免疫排斥反应的相关研究奠定基础,也为下一步进行基因沉默、基因修饰和基因敲除提供基因图谱框架,确定研究的重要目标靶基因,最终为版纳微型猪近交系在猪→人异种器官移植领域中的研究和应用奠定基础。 Banna Mini-pig Inbred Line (BMI) is the first successful cultivated large mammal inbred line in the world. It's one of the major scientific and technological achievements with independent intellectual property rights which is innovated by Chinese primitively. The great research and application value can be found in the bio-medical field such as xenotransplantation. This research intends to explore all the genes relating to the immunological rejection in the xenotransplantation of BMI to human based on transcriptome sequencing technology (including unknown and rare transcription). Analysis the obtained data by functional enrichment analysis and cluster analysis to screen the major genes associated with xenotransplantation rejection. In order to determine the genes with high level expression in major organs for xenotransplantation, we will detect multiple tissue expression in transcription levels by real-time qRT-PCR. Then the function will be identified by prokaryotic and eukaryotic cells in protein express level. Finally, identified the main exclusive genes related to major transplanted organs in the DNA-RNA-protein level. All of these research prepared the ground for studying the genes correlated with immunological rejection in the xenotransplantation of pig to human. And it also can provide the gene mapping framework for gene silencing, gene modification and gene knock as well as detect the important target genes further. Finally it can lay the foundation for the BMI research and application in the xenotransplantation of pig to human field. |

It could therefore lay the foundation for the BMI research and application in the xenotransplantation of pig to human field. |
