Poly (ADP-ribose) polymerase(PARP-1 and PARP-2) are the key enzymes responded to the DNA damage by the process of base excision DNA repair (BER) pathway to maintain the life circle of cell survive and proliferation. It has been reported that PARP inhibitors could potentiate the antitumor effects of chemotherapies and radiation therapy. Most importantly, PARP inhibitors could selectively kill cancer cells with BRCA (BRCA-1 and BRCA-2) defects which are lack of homologous recombination (HR) pathway of double-stranded DNA repair. These research results make PARP to be a potentially important therapeutic target for cancer patients. |
