糖苷类香气前体过柱问题

About 1 g of fresh tea leaves, an appropriate amount of quartz grain and liquid nitrogen were simultaneously placed in a mortar. When the leaves were ground to emulsion, phenyl beta-D-glucopyranoside and polyvinyl polypyrrolidone (PVPP) were added, and the samples were further ground to enable their mixing. This emulsion was transferred into a 50 mL centrifuge tube, and subsequently ultrasonically extractionfor 20 min using 15 mL methanol, centrifuged for 10 min at 10,000 rpm. The residue was re-extracted two times using methanol (15 mL *2), and all the supernatants were combined and evaporated to dryness with nitrogen. "The dried sample was re-dissolved using 2 mL water, of which 0.2 mL was subjected to the C18 cartridge. 20 mL of methanol was used for leaching solution, and the effluents were collected and evaporated to dryness with nitrogen, and further dehydrated in a lyophilizer for 24 h.” 我觉得引号里的那段不太对,照着做了,用真空一抽,再用甲醇洗脱,没用啊,本来用的就是500mg、6CC的一次性C18固相萃取小柱,文章里都没有写活化柱子,淋洗不要的东西之类的,故意的?各位大侠,有人做糖苷的吗?迷茫啊,不会过柱,就算最后色谱图跑得很好,没有标准品也不知道它是什么东西啊?对了,这篇文献是“GC–ECD Method for Determination of Glucosidically Bound Aroma Precursors in Fresh Tea Leaves”。 |
