如何理解stability-indicating

在药品稳定性考察中，要求使用指示稳定性的分析方法(stability-indicating analytical method). 那么我们应该如何理解stability-indicating 呢？我在网上看这样一段话FDA defines stability-indicating assay as an analytical method that accurately quantitates the active ingredients without interference from degradation products, process impurities, excipients or other potential impurities。从这段话看FDA将指示稳定性的分析方法定义为能够准确定量分析活性成分而不受降解产物、工艺杂质、辅料或其他潜在杂质干扰的分析方法。那么请问大家是如何理解stability-indicating的呢？哪些方法需要是指示稳定性的？又如何证明分析方法指示稳定性的能力的？ [ Last edited by yan1302 on 2006-12-25 at 08:44 ] |

原料药（或成品）在制备和贮存期间可能分解，在建立规格以及检测方法时应该有所考虑。基于许多药物本身的复杂性，并无单一的的稳定性指标分析或参数可描绘全部药物或成品的稳定性。因此，应根据药物本身拟定稳定性指标分析方法，用以探讨药物品质的变化，需采用哪些测试则取决于药物本身的特性。 在药物研究的早期开展稳定性苛刻实验是受鼓励的，因为这些研究提供的资料对实验选择稳定性实验分析方法是相当重要的。强迫降解实验是分析方法的stability-indicating验证的一个重要部分。理论上来讲，稳定性实验检验方法无论是否与成品放行检验方法一致，都必须进行强迫降解实验，以证明该方法是stability-indicating。既便是药典方法，也必须这样做。（如做：药物的影响因素实验分析，比如强酸强碱破坏实验等，主要是考察药物在不同条件下的稳定情况） 可以参考《Stability-Indicating HPLC Methods for Drug Analysis》（可惜我没有这本书，哪位有的，能否共享啊？） [ Last edited by 广之 on 2006-12-24 at 22:40 ] |

感谢3楼的回答。FDA给台湾Delta合成的警告信有关稳定性试验的原文是这样的： Stability-indicating methods were not used in the stability testing program for [ ] You response indicates that you will use stability indicating methods in the future stability testing of [ ]. However, it fails to indicate whether you are currently using stability indicating methods in your stability program and did not provide a timeframe for beginning the use of stability indicating methods. The test procedures used in stability testing should be validated and be stability indicating. please provide documentary evidence that you are currently using stability indicating methods for all stability testing of [ ] and any other API manufactured for US market. 另外，分享一篇有关该主题的文章。 [ Last edited by yan1302 on 2006-12-25 at 08:46 ] |
