| *** is the world's first successful cultivation of a large mammal inbred,playing a important role in carrying out xenotransplantation, animal models of disease, transgenic animals, genetically modified, gene knockout and functional genomics research .This research focuses on new genes related to immune rejection during *** xenotransplantation Process , through the use of digital gene expression profiling method £¬the high-throughput screening gene and illumina sequencing were carried out, the sequencing results was evaluated, gene annotation, EST comparison, the new gene associated with immune rejection was screened and determined preliminary. Expression of Gene was detected by the means of real-time PCR and Northern-blotting,the key gene associated with immune rejection was determined through correlation analysis of expression level and immune rejection£¬and multi-tissue expression pattern analysis, function prediction and identification were carried out.It is not only lay the foundation for revealing xenograft rejection in immune research£¬but also for identifying further important target gene in conduct of genetic modification and gene knockout ,finally it provides a scientific basis for increase of the success rate in the field of xenotransplantation. |
