求助，这个研究计划应该怎么写？？ 已有1人参与

求助各位大侠，老师让写个研究计划，可是无处下手啊，之前不是学分子生物学的，请各位给一点建议，研究计划的提纲如下： Regarding to your application, can you write a research proposal relevant to Wnt pathway? I would propose the following topic for your research proposal: Developing cell-based assay for Wnt signaling. The outline will be as followed: 1. Construct a vector encoding Frizzled receptor (a GPCR). Please include the detailed information of choosing the vetor, designing the primers for PCR and the whole cloning process, etc. 2. Construct a vector encoding report gene containing TCF-LEF responsive element (luciferase report gene or GFP gene) Please include the detailed information of choosing the vetor, designing the primers for PCR and the whole cloning process, etc. 3. Generate a cell line stably expressing the above two genes. Please include the process of stable cell line generation. 4. Expressing Wnt protein in bacteria expression system with affinity tag (His tag or GST tag) and purification of Wnt proteins. Please include the detailed information of choosing the vetor, designing the primers for PCR, the whole cloning process and protein purification, etc. 5. Verify the cell-based assay and carry out the protein activation assay During the writing process, you could refer to the webpage http://www.stanford.edu/group/nusselab/cgi-bin/wnt/ and check out the relevant literature. 万分感激！！ |

怎么会无处下手？ 以第一个为例 事情：建载体 目的基因：Frizzled receptor (a GPCR) 什么物种？ 人物：你自己，可问师兄师姐 过程：什么叫Frizzled receptor (a GPCR)， 得查文献，用www.google.com查，或者学校数据库，实在不懂的话，就先看中文的理解，后再看英文的。 研究计划撰写：1 为什么要建该载体，意义何在 2 关于连在载体上该基因的相关研究进展 3 如何建， 别人问献可以参考。 要记住：如果什么都可以用教来解决的话，那就不叫研究，顶多叫高中生生实验。 |
