| A method for the prepn. of Sucralose was designed. A reaction of sucrose with trityl chloride and acetylation provided TRISPA [i.e., 2,3,3',4,4'-penta-O-acetyl-1',6,6'-tri-O-tritylsucrose] which was deprotected to deliver 3,4-di-O-acetyl--(D-fructofuranosyl)--D-glucopyranoside 2,3,4-triacetate (4-PAS). This intermediate was transformed into 3,4-di-O-acetyl--(D-fructofuranosyl)--D-glucopyranoside 2,3,6-triacetate (6-PAS). Chlorination of the above-mentioned intermediate using triphenylphosphine and thionyl chloride provided sucralose pentaacetate. The byproduct (triphenylphosphine oxide) was reclaimed. The synthesis of the target compd. (Sucralose) was achieved after deacetylation. This method is simple, byproducts are easily reclaimed and starting materials are inexpensive. |
