土壤中微生物和多环芳烃(PAHs)降解功能基因群落多样性的研究对PAHs污染土壤的微生物修复具有重要指导意义。土壤中PAHs降解功能基因和PAHs降解的标志微生物含量高低和多样性极大地影响了能否在土壤中筛选到PAHs高效降解菌株和能否利用生物刺激降解土壤中的PAHs。近年来,分子生物学的发展极大地扩展了我们对于微生物多样性的认识。分子生物学方法可获得关于土壤微生物群落的更精细结构信息,可更为全面的反映微生物区系组成状况。本研究以某焦化厂场地土壤为研究对象,通过分析土壤PAHs含量和土壤理化性质,并利用土壤DNA提取、普通PCR、实时定量PCR和T-RFLP等现代分子生物学手段研究土壤细菌的丰度和群落多样性,探究土壤微生物丰度和群落多样性变化的影响因素。在此基础上,以深层PAHs污染土壤为对象,通过振荡措施改善土壤供氧速率,考察供氧能否促进深层土壤PAHs降解以及降解过程中土壤微生物丰度和细菌群落多样性动态演变规律。

Study of soil microorganism and diversification of PAHs degradative functional gene community is of great instructive value to microbiological bioremediation of PAHs polluted soil. Contents and diverification of soil PAHs degradative functional genes and PAHs degradation featured microorganisms greatly determine isolation of PAHs degradative strains with high efficiencies, and bioremediation of PAHs polluted soils. Recently, molecular biology greatly broadens our knowlege on diversification of soil microorganisms. With molecular biology, we can get detailed information on community structures of soil microorganisms, as well as floristic compositions of microorganisms. Starting from polluted soils, sampled from a coking plant, aboundance and diversification of soil microorganisms were investigated through analysis of physiochemical properties and PAHs contents of polluted soil. Molecular techniques, such as soil DNA isolation, PCR, realtime PCR and T-RFLP were also applicated. Influential factors of aboundance of soil microorganisms and community diversification were discussed. With PAHs polluted deep soil,soil oxygen exchanging rate was improved by shake to investigate whether it can help degradate PAHs from deep soil. Meanwhile, evolution features of aboundance of soil microorganisms as well as dynamic community diverisification structures were investigated.
