构建pSos-MLPKf2诱饵质粒,将诱饵质粒转化入酵母感受态中,酵母生长情况表明,该诱饵对酵母细胞生长无毒性。同时自激活检验结果表明该诱饵无自激活作用,所以可以在CytoTrap酵母双杂交系统中用于筛选实验。诱饵pSos-MLPKf2质粒与文库质粒共转化酵母感受态,筛选效率达到104,符合试验要求。经过多次重复筛选,获得1个阳性克隆,测序后经BLAST分析表明插入序列是SHV3的同源序列,同源性达到77%。这为更加深入的研究MLPKf2的功能和SI分子机制的研究提供了新的线索。

PSos-MLPKf2 bait plasmid was constructed and transformed into competent yeast. The results of yeast growth showed that the bait hadn’t toxicity on the yeast cell growth. This bait could be used for screening experiments in the CytoTrap yeast two-hybrid system for the activation of the test showed that the bait had no self-activation. The filter efficiency of pSos-MLPKf2 bait plasmid and library plasmids were transformed into yeast competent was 104, which meet the test requirements. A positive clone was obtained after repeating screening. The sequenced tested by BLAST showed that the inserted sequence is the homologous sequence of SHV3 and the homology was 77%. The research provided a new clues for more in-depth research of MLPKf2 function and molecular mechanism of SI.
