The cells are washed in cold sterile distilled water and dehydrated by precipitating with three volumes of acetone at –20°C, and then they are allowed to stand at –20°C for 24–48 hours. After repeated washing in cold acetone, followed by a final rinse in diethyl ether, the cells are dried over calcium chloride and held at 4°C. The dried cells are subjected to a viability check. They are resuspended in sterile 2.5% sodium chloride to a final concentration of 5% (w/v) and agitated for 3 days at 4°C. |
