做免疫胶体金时看到的一个小册子，最近常在参考文献中出现。托US的朋友描的。希望有用： Beesley, J. (1989). "Colloidal Gold. A new perspective for cytochemical marking." Royal Microscopical Society Handbook No 17; Oxford University Press: Oxford, U.K. 顺便问问大家测胶体金的PH用什么测的，PH试纸还是PH计？不知有没有高人用过gel-filled的PH电极？现在我的问题是这样的，测胶体金溶液时，用PH试纸和PH计（上海）会相差2个PH单位。很可能是这个普通PH计被堵掉了。早期的经典文献中建议测胶体金时用gel-filled的PH电极。他们是如此描述的： 1.Beesley, J. (1989) ：The pH ofthe colloid is an important factor in these reactions. It is inadvisable to use pH paper for estimating the pH of the sol because this method is not sufficiently accurate. A pH electrode is the preferred equipment but the gold sol will plug the pores unless suitable precautions are taken. Electrode damage is prevented by the addition of a few drops of 1 per cent aqueous polyethylene glycol (MW 20000) or carbowax-M to a few millilitres of the colloid before measuring the pH (Lucocq and Roth 1985). This stabilized colloid must be discarded after the pH measurement is taken. Alternatively, a gel-filled combination electrode may be used, in which case contact between the gold sol and the electrode should be kept to a minimum. 2.Geoghegan, W. and G. Ackerman (1977).：To check the pH of colloidal gold two drops of 1% polyethylene glycol (PEG) MW. 20,000 were added to 10 ml of colloidal gold to stabilize it before the pH electrode was inserted. Failure to stabilize the gold before measuring pH results in the porus plug or wick of the electrode becoming plugged with colloidal gold, presumably flocculated by the KC1 in the electrode (we found estimates made with pH paper to vary by as much as 0.5 units). 3. 可以下了。 gel-filled应该不是一般的电极吧，要不THERMO收购的奥立龙专门有GEL-FILLED的产品http://www.thermo.com.cn/Search.aspx?keyword=gel%20filled和SURE-FLOW的专利产品呢。哈希也有，我是在GOOGLE中搜了才知道的。正在和THERMO客服问询。呵呵。 |
