We use five different kinds of commercial protease to hydrolyze shark skin gelatin, and measure the antioxidant activity index of hydrolysis products such as their abilities to eliminate DPPH•radicals and ABTS radicals, as well as their inhibition to Fe(II)-induced yolk lipoprotein polyunsaturated fatty acid peroxidation. The result shows that acidic protease hydrolysis products have the best antioxidant activity. Furthermore, we use SP-Sephadex C-25 cation exchange chromatography and Sephadex G-50 gel filtration chromatography to separate the acidic protease hydrolysis products and get an strong elution peak MS to eliminate the activities of DPPH•radicals. And Tricine-SDS-PAGE experiment indicates that their molecular weight distribution is smaller than 3.3kD. |
