分别使用5种不同的商业蛋白酶对鲨鱼皮明胶进行水解，并测定其水解产物的DPPH•自由基清除能力、ABTS自由基清除活性以及对Fe2+诱导卵黄脂蛋白多不饱和脂肪酸过氧化反应的抑制作用等抗氧化活性指标。结果表明，酸性蛋白酶水解产物具有最佳的抗氧化活性。再利用SP-Sephadex C-25阳离子交换色谱和Sephadex G-50凝胶过滤色谱对酸性蛋白酶水解产物进行分离，得到具有较强的清除DPPH•自由基活性的洗脱峰M5。Tricine-SDS-PAGE试验表明其分子量分布小于3.3KD。 [ Last edited by asd123xukun on 2010-7-28 at 21:19 ] |

| We use five different kinds of commercial protease to hydrolyze shark skin gelatin, and measure the antioxidant activity index of hydrolysis products such as their abilities to eliminate DPPH•radicals and ABTS radicals, as well as their inhibition to Fe(II)-induced yolk lipoprotein polyunsaturated fatty acid peroxidation. The result shows that acidic protease hydrolysis products have the best antioxidant activity. Furthermore, we use SP-Sephadex C-25 cation exchange chromatography and Sephadex G-50 gel filtration chromatography to separate the acidic protease hydrolysis products and get an strong elution peak MS to eliminate the activities of DPPH•radicals. And Tricine-SDS-PAGE experiment indicates that their molecular weight distribution is smaller than 3.3kD. |
