| 温阳方由温阳利水法化裁而来，是王老经长期临床实践总结的慢性心衰中药复方制剂。前期临床和实验研究证实该方具有良好的临床疗效，能够改善心力衰竭动物模型心肌重塑。随着慢性心力衰竭研究进展的不断深入，细胞因子网络在心力衰竭心肌重构中的作用日益引起广泛关注。其中肿瘤坏死因子α(Tumor necrosis factorα，TNFα) 是重要的促炎性细胞因子，作为激活细胞级联反应的主要介质，在细胞因子网络中发挥关键作用。核转录因子-κB (nuclear factor κB，NF-κB)是调控多种基因转录的关键因子，活化后能调控包括TNFα在内的多种炎症介质的转录，作为炎性反应的整合者在免疫调节和炎症发生过程中具有重要作用。为进一步探讨温阳方药效和作用机理，明确NF-κB在心衰发病中的作用及对TNFα调节机制，本研究采用细胞培养和血清药理学方法，开展温阳方对肿瘤坏死因子α（TNFα）诱导新生大鼠心肌成纤维细胞（cardial fibroblasts CFs）增殖、胶原合成，及核转录因子NF-κB表达的影响，进一步从分子水平探讨其抑制心肌纤维化，改善心力衰竭心肌重塑的作用机制，从而为其临床应用及新药开发提供科学理论和依据。 |

Transcription factor - κ B (nuclear κ B, NF - in κ B) is the regulation of gene transcription factors, activate the key to control including TNF alpha, inflammation, as the transcription of integration of the inflammatory response in immune adjustment and inflammation plays an important role in the process. For further discussion WenYangFang efficacy and mechanism, clear in heart failure κ B NF - the role and pathogenesis of TNF alpha adjusting mechanism, this study adopts cell culture and serum pharmacology, WenYangFang method of tumor necrosis factor (TNF alpha) inducing newborn rat cardiac fibroblasts cardial (CFs) proliferation, fibroblasts collagen synthesis, and κ transcription factor NF - expression of B, further discusses its suppression of the molecular myocardial fibrosis, improve cardiac remodeling of heart failure mechanism, so as to its clinical application and development of new medicine to provide scientific theory and evidence. |
