| Abstract: In order to study the toxicity of fluoride on rat¡¯s kidney tissue, the polluted water containing 15, 30, and 60mg /L of sodium fluoride was fed to rats in a natural way. The rats were put to death after 9 months. The oxidation level and nuclear factor ¦ÊB (NF-¦ÊB) expression level in rat¡¯s kidney were determined. The results showed a visible morphological change among each group of fluoride poisoned rat¡¯s kidneys and the body weights of high dosed group were lower greatly than the controlled group. Both MDA contents of low dosed and high dosed groups were higher than the controlled group. The enzyme activities of GSH-Px of low dosed group and the high dosed group were obviously higher than the controlled group. Although there were increasing trends of SOD activity, the difference is not considerable. NF-¦ÊB expression level was rising as the exposure dose increased and that of high dosed group was dramatically higher than the controlled group. The study indicated that Fluoride could decrease the activity of antioxidant enzymes in rat¡¯s kidney and increase lipid peroxidation and NF-¦ÊB expression level, which caused the damage of kidney tissue. NF-¦ÊB in kidney is sensitive to fluorosis, which can be taken as one of molecular diagnostic markers for fluoride damaged kidneys. |

all rats exposed to fluoride showed visible histopathological renal morphological changes, high-dose group body weight was lower obviously(P<0.05), the MDA contents of low-dose, high-dose groups were obviously increased(P<0.05). The enzyme of GSH-Px activity of low-dose group obviously higher than the control group (P<0.05), high dose group increased significantly (P<0.01). As to SOD activity, although a downward trend, but no obviously difference. As the exposure dose increased, NF-¦ÊB expression level was also on the rise, high-dose group obviously higher than the control group (P<0.05). The results indicated that chronic fluorosis can reduce the activity of kidney antioxidant enzymes in rat, resulting in an increase in lipid peroxidation, NF-¦ÊB expression level increased, caused kidney tissue damage. NF-¦ÊB in kidney tissue is sensitive to fluorosis, which can be one of molecular diagnostic makers for kidney damage by fluoride. ÕâÒ»¶Î¿´´óϺÃÇ»¹Äܲ»ÄÜÔٸĸģ¬Ç°Ã涨ÏÂÀ´²»¶¯ÁË¡£ |
