The use of dry weight measurements for quantification of the stock solutions is a crude and in-accurate means of protein quantification. Often times in commercially purchased peptides greater than 10% of the dry weight is constituted from salts and residual water (the % purity values indicated by the manufacturers relate to what % of all peptide material in the vial is the target peptide - it does not indicate what percent of all dry weight is the target peptide). 我是用标准物质配置标准溶液建立标准曲线 然后对粗蛋白质样品中的这个标准物质进行定量 1.审稿人为什么说我是标准溶液的定量啊？ 2.无论样品组成如何，只要是我建立的串联质谱方法合适，就可以进定量啊 和杂质无关啊 |

The use of dry weight measurements for quantification of the stock solutions is a crude and in-accurate means of protein quantification. 称干物质配置标准比较粗略，在蛋白质定量中不精确 Often times in commercially purchased peptides greater than 10% of the dry weight is constituted from salts and residual water 通常购买的PEp中超过10%的是盐和水分 (the % purity values indicated by the manufacturers relate to what % of all peptide material in the vial is the target peptide - it does not indicate what percent of all dry weight is the target peptide). 商品的纯度指的是所有peptide中含所含有目的pep的百分比（target peptide ），而不是净重中含有目的pep的百分比。 净重* X% 要大于目的pep含量。因为peptide种类也较多吧，呵呵  就像 氯化钠氯化钾冰棍（氯化钠纯度98%，应该是指占总盐的比例），你要计算食盐的含量，你要除去水，还要除去氯化钾 [ Last edited by bear2007 on 2010-3-24 at 14:13 ] |
