2.2. Preparation of dispersions b-lg-Enriched powder was dispersed (6%, w/w) in distilled/deionised water or in a 100mM NaCl solution with stirring at room temperature. b-lg-Stabilised emulsions were prepared for zeta potential determination. A pre-emulsion of 1% b-lg and 20% rapeseed oil (Spar Ltd., Dublin 12, Ireland) was prepared at pH values of 7.0 and 3.0 using a using a Silverson L4RT mixer (Silverson Machines Ltd., Waterside, Chesham, Bucks, England) for 10 min at 5000 rpm. The mixture was homogenised using a Microfluidizers M-110 EH Processor (Microfluidiser Corporation, 68623 Lampertheim, Germany) at 22 1C and 1500 bar. The emulsions formed at pH 7.0 and 3.0 had particle size values (D4,3 values) of 282724 and 9737106 nm, respectively. Emulsions with intermediate pH values were prepared by combining the emulsions prepared at pH 7.0 and 3.0 in various ratios. Chitosan (1%, w/w) was dissolved by dispersing in water at 22 1C by adjusting the pH to 3.0 using 1M HCl. Subsequent pH adjustment was performed with 1M NaOH. Mixtures of 1% (w/w) b-lg in water or 100mM NaCl, or 1% (w/w) b-lg and 0.1% (w/w) chitosan in water or 100mM NaCl were prepared by combining stock solutions (6%, w/w, b-lg; 1%, w/w, chitosan; 500mM NaCl; water) at pH values ranging from 3.0 to 8.0. |
