Nine genes were found differentially expressed between days 1 and 3 of culture. These genes included the transcription factors peroxisome proliferator-activated receptor gamma 1 and 2 (PPARG1 and PPARG2) which showed an increased expression during proliferation. Similarly, an increased expression was observed for genes encoding proteins involved in fatty acid transport and activation such as solute carrier family 27 (FA transporter) member 1 (SLC27A1) and short-chain acyl-CoA synthetase 2 (ACSS2). The expression of the fatty acid oxidation genes catalase (CAT) and carnitine O-palmitoyltransferase 2 (CPT2) were also induced, whereas, the expression of carnitine O-palmitoyltransferase 1A (CPT1A) was reduced during cell proliferation. Moreover, the expression of fatty acid desaturase 2 (FADS2) and the adipocyte secreted factor leptin (OB, also known as LEP) increased during proliferation. Preadipocyte differentiationwas induced at day 3 of culture. This led to altered expression of a majority of the genes studied. Only long-chain acyl-CoA synthetase 3 (ACSL3), carnitine O-palmitoyltransferase 1B (CPT1B), peroxisome proliferator-activated receptor beta (PPARB), and solute carrier family 27 (FA transporter) member 5 (SLC27A5) did not show any changes in expression during preadipocyte differentiation. |
