In this experiment, assessment of the metabolic activity of differentiating preadipocytes confirmed gene expression, showing that preadipocytes almost exclusively stored FAs in the form of TAG. In fact, only 1.3% of the [14C] 16:0 taken up by the cells was utilised for energy through ¦Â-oxidation and the remaining 98.7% was stored in the cells. Due to the low total oxidation in the differentiating preadipocytes, analysis of ASPs by HPLC did not give conclusive results on oxidation end products. Moreover, 82.2% of the stored [14C] 16:0 was recovered in TAG, whereas the remaining 16.5% was recovered in PL. Only small immeasurable amounts of [14C] 16:0 could be found in the remaining lipid classes. In order to investigate the degree of FA modifications present during late adipogenesis the TAG and PL fractions were separated by silver ion TLC. Interestingly, 54% of the radioactive 16:0 had been desaturated and elongated into 18:1n−9 in both TAG and PL fractions .
