发酵液中黑色素提取的问题

我培养了一种产黑色素的酵母,有时产黑色素,有时不产,我也不知道产黑色素的条件是什么,可我觉得这黑色素可能有一定的研究价值,请问用什么方法能把黑色素提出来呢?我的黑色素是水溶性的。 [ Last edited by 350784478 on 2009-10-17 at 17:05 ] |

请参考高产黑色素菌株的筛选及其发酵条件与黑色素结构的初步研究 -------------------------------------------------------------------------------- 论文标题:高产黑色素菌株的筛选及其发酵条件与黑色素结构的初步研究 The Screening of the High-Melanin-Producing Strains and Studies on the Fermentation and the Sturcture of the Melanin 论文作者 论文导师 万波,论文学位 硕士,论文专业 微生物学 论文单位 中国科学院研究生院(成都生物研究所),点击次数 11,论文页数 55页File Size2812k 2005-06-01论文网 http://www.lw23.com/lunwen_732675287/ melanin; streptomyces; identification; structure; HPLC; MS 对从土壤中分离出的菌种进行筛选,从中筛选出产胞外黑色素菌种27株,其中高产黑色素菌种4株,并对其中的88号菌种进行了发酵条件的研究,确定了高产黑色素的培养基及培养条件。对88号菌种所产黑色素的结构进行了初步鉴定,确定其黑色素由11种成分组成,并推测出其中8种成分的分子式。 对从土壤中分离出的321株菌株进行了筛选,得到4株高产胞外黑色素的菌株。通过比较了其在不同培养基上产黑色素的能力,确定88号菌作为高产黑色菌种。经形态和生理生化鉴定,初步确定该菌株为链霉菌属。鉴于该菌黑包素产量高,可以作为一类新的产黑色素菌种资源。 设计了一系列实验,研究了pH值、钠离子、镁离子、氨、蛋白胨、葡萄糖、酪氨酸等对黑色素产量的影响,确定了其黑色素产率最高的培养基,并对其发酵条件进行了优化,确定了最佳的发酵条件。 从88号菌种发酵液中分离提取出了黑色素,并通过萃取、薄层层析、硅胶柱层析、高效液相色谱等方法对黑色素进行了纯化,对纯化后的黑色素进行了ESI-MS检测,确定其主要由11种成分组成,并对这11种成分进行了高分辨率质谱检测,结合已有的相关文献对所得黑色素结构进行了分析,推测出其中8种成分的分子式。 The strains isolated from the soil were screened according the ability of melanin-producing and 27 strains were acquired among which there were 4 high-melanin-production strains. The fermentation condition of the strain No.88 was studied , the culture medium and the culture condition were confirmed. The melanin produced by the strain No.88 was concluded to be consisted of 11 components according to the preliminary study of this melanin and 8 of whose molecule formula were speculated.4 strains who produce extracellular melanin were acquired after the screening among the 321 strains isolated from the soil and strain No.88 was confirmed as the high-melanin-producing strain according the ability of melanin-producing on different culture mediums. The strain No.88 was confirmed to be streptomyces according the morphological, physiological and chemical characters. Because of its high-melanin-production, the strain No.88 may be a new kind of strain source in the melanin-producing microbe.A series of experiments were designed to study the effect of the pH, Na~+, Mg~+, (NH_4)~+, peptone, glucose, tyrosine to themelanin-production. The best culture medium was determined and the best culture condition was confirmed after the culture condition optimizing.The melanin was isolated from the fermentation mixture of the strain No.88. After purification by the means of extraction, folium separation, silica gel column separation, HPLC, the ESI-MS detect was hold and the melanin was determined to be consist of 11 components and the HR-MS was hold to detect each of those components. The molecular formulas of 8 of those components were speculated according the correlative literatures on the structure of the natural melanin. |
