Prior work in the Zhu lab has determined that Sirt5 decreases in cartilage during aging and is an important regulator of chondrocyte cellular metabolism (Zhu, et al. Cartilage, 2021). It increases activity of hundreds of metabolic proteins in chondrocytes through removing post-translational malonylation (MaK) on the lysine sites. We have also demonstrated that MaK is increased in obesity (Liu, et al. Osteoarthritis and Cartilage, 2022) and has an intriguing link with de novo lipogenesis in chondrocytes (Liu, et al. JOR, 2022).
Our goal is to understand the role of Sirt5-MaK in chondrocyte cellular metabolism and osteoarthritis development during aging and obesity. Fundamental knowledge gained from this understanding will be leveraged to develop novel therapeutic approaches to treat degenerative pathology in murine models of osteoarthritis. We will employ novel stains of transgenic mice with conditional modulation of Sirt5-MaK in articular cartilage and apply state-of-the-art techniques such as metabolomics, single cell RNA-Seq, and Seahorse Respirometry.
