参考一下以下内容 Following hypotonie rupture of the cells with glass-distilled water and dilution (1:1) with 300 miviKC1-100 min Tris buffer (double strength KCl-Tris), the microsomal fractions from both the sensitive and the drug-resistant cells were obtained by differential centrifugation in KCl-Tris, as previously described ( 17). The mitochon-drial fractions were isolated by centrifugation in 0.25 Msucrose-25 mM Tris, pH 7.4, containing 1 mM EDTA and 1% BSA, followed by washing three times in KCl-Tris buffer, as previously described (18). The purified mitochondria and microsomes were suspended in KCl- Tris buffer and diluted appropriately following the determination of the protein content. |
